FAQ.

Reposition the QuarterBento (top left corner) in its location and engage its four clips at the periphery by gently pressing a plug on the QuarterBento. For sterility reasons, always handle the QuarterBento with gloves and under a flow hood.
Remove the vacuum bag, place the lid on the NeoBento and place it in a clean vacuum dessicator for 30-45 min before processing to chip preparation.
Verify the level of water into the labyrinth, both in the NeoBento frame and the plug. Remember that you have to fill up below the height of the shorter labyrinth walls.
The concentrations mentioned in the Operating Protocol are indicatives. They were determined after counting cells with a Septer (Merck Millipore - diameter size: 7-16 µm) but may vary depending on the counting technique. If you want to adjust the concentrations for future experiments, perform seeding tests with different increasing cell concentrations to establish the optimal concentration for your experiments.
Neurite extension may be difficult to observe in transmitted light over the microchannels. If you have the possibility to stain the cells, check with a fluorescent stain for the absence or presence of neurites.
Remove the bottom cover and verify the absence of humidity on the bottom of the chips and/or the surface of the bottom cover.
